hMSCs(human marrow mesenchymal tem cells) were obtained from Prof. Lingsong Li(Peking University Stem cell Research Center,Beijing China).This is a well-characterize hMSC cell line. hMACs were cultured with 10% FBS in Alpha MEM complete media and were plated in a 25-cm2 flasks at a density of 10^4 cells/cm2 or into wells of 24-well plate at a density of 3000~4000 cells/cm2 and incubated at 37℃ in 5% CO2. After 72 hours, the meida were replace. At 80%~90% confluence, cells were harvested with 0.25% trypsin(Sigma) for 3 minutes at 37℃. Osteogenic differentiation was assessed by incubating hMSCs with Alpha MEM osteogenic-inducing media for 7,14 or 21 days with media changes twice weekly.

Dopamine neurons derived from embryonic stem cells function in an animal model of Parkinson's disease
Parkinson's disease is a widespread condition caused by the loss of midbrain neurons that synthesize the neurotransmitter dopamine. Cells derived from the fetal midbrain can modify the course of the disease, but they are an inadequate source of dopamine-synthesizing neurons because their ability to generate these neurons is unstable. In contrast, embryonic stem (ES) cells proliferate extensively and can generate dopamine neurons. If ES cells are to become the basis for cell therapies, we must develop methods of enriching for the cell of interest and demonstrate that these cells show functions that will assist in treating the disease. Here we show that a highly enriched population of midbrain neural stem cells can be derived from mouse ES cells. The dopamine neurons generated by these stem cells show electrophysiological and behavioral properties expected of neurons from the midbrain. Our results encourage the use of ES cells in cell-replacement therapy for Parkinson's disease.
